I hope you meant "line up 4 stones", since otherwise it's just as
trivial as on a 3x3 board. More trivial, in fact, as the first
player can always win in 5 turns by starting in one of the center
squares.

Not that 4-in-a-row would be much harder - an exhaustive search
can find an optimal strategy in no time.
